<i>De Novo</i>-Designed β-Sheet Heme Proteins

The field of de novo protein design has met with considerable success over the past few decades. Heme, a cofactor, often been introduced to impart diverse array functions protein, ranging from electron transport respiration. In nature, heme is found occur predominantly in α-helical structures β-sheets, which resulted significant designs proteins utilizing coiled-coil helices. By contrast, there are only known β-sheet that bind and β-sheets frequently result amyloid-like aggregates. This review reflects on our designing series multistranded binding peptides well folded both aqueous membrane-like environments. Initially, we designed β-hairpin peptide self-assembles performs peroxidase activity membrane. was optimized further accommodate pocket within for catalysis transfer membranes. Furthermore, characterized miniproteins soluble an environment capable single multiple hemes high affinity stability. Collectively, these studies highlight substantial progress made toward functional β-sheets.
